Design and development of loteprednol etabonate nanodispersion for ophthalmic drug delivery

Postoperative inflammation after cataract surgery can prolong visual recovery time, increase the treatment burden and elevate risk of poor outcomes. Corticosteroids are potent anti-inflammatory agents that have been used successfully for decades to treat ocular inflammation. However, side effects associated with topical corticosteroids including increased intraocular pressure, formation long-term use, decreased resistance infection major concerns. Amongst available Loteprednol Etabonate (LE), an ester-based corticosteroid has selected studies. The aim present study was develop etabonate loaded nanodispersion steroid responsive inflammatory conditions eye objectives reducing frequency administration increasing solubility drug. Non-solvent method formulate nanodispersion. formulation assessed particle size polydispersity index, drug content, surface morphology, isotonicity, test sterility, in-vitro & ex-vivo release. Mean PI optimized batch found be 50±10nm 0.2±0.1. No microbial growth observed in during sterility testing isotonic when compared marketed plain blood cells.
